History and culture
N-ethylhexedrone was first identified in a sample from the Belgian customs laboratory received by the JRC in November 2015. In January 2016, it was identified at JRC in a sample provided by French customs. Subsequently, in February 2016, EMCDDA received reports of the identification of this substance from other countries, such as Sweden, the Netherlands, France, Belgium and Slovenia.
Chemistry
n-Ethylhexedrone is a molecule from the chemical class of cathinones. The term “substituted cathinone” refers to a wide range of substances based on cathinone, the main active component of the khat plant. Cathinone essentially consists of an amphetamine nucleus (a phenethylamine nucleus with an alkyl group attached to the alpha carbon atom) and an oxygen group attached to the beta carbon atom. Cathinones are also known as beta-ketone (βk) analogues (double-bonded oxygen at the β-carbon atom) of amphetamines.
In particular, the cathinone backbone can be modified at three different sites, creating hundreds of possible compounds, including substituents such as those
on the aromatic ring, the alpha carbon, or the amine group.
pharmacology
Very few data are available on the pharmacokinetics and pharmacodynamics of n-ethylhexedrone and other substituted cathinones in humans. Like amphetamines, synthetic cathinones exert their stimulatory and sympathomimetic effects via increasing synaptic concentrations of catecholamines such as dopamine, serotonin, and norepinephrine. These molecules are able to inhibit monoamine reuptake transporters, resulting in reduced clearance of neurotransmitters from the synapse. They can also cause the release of biogenic amines from intracellular stores.
Synthetic cathinones are generally less able to cross the blood-brain barrier than amphetamines because the beta-keto group causes increased polarity. Unlike other synthetic cathinones, pyrrolidine derivatives have a higher ability to cross the blood-brain barrier because the pyrrolidine ring gives these molecules a low polarity. Studies on the metabolism of synthetic cathinones have shown that they are n-demethylated, the keto group is reduced to hydroxyl and the ring alkyl groups are oxidized.
